Smad2/3 bind to Smad4, translocate to the nucleus and activate, among others, genes that directly regulate EMT. Smad7, negatively controls the TGF-β signaling by inhibiting R-Smads phosphorylation, by competing directly with Smad2 and 3 for the TGF-β-Receptor binding.
