Gill kreeg in New York privé-onderwijs. Al op jonge leeftijd had hij interesse in [[natuurlijke historie]]. Hij was betrokken bij [[J. Carson Brevoort]]s voorbereidingen voor diens entomologisch een ichtylogische verzamelingen. In 1863 vertrok Gill naar [[Washington D.C.]] om te gaan werken bij het [[Smithsonian Institution]]. Hij catalogiseerde onder andere [[zoogdieren]], [[vissen (dieren)|vissen]] en [[weekdieren]]. Tevens was hij bij Smithsonian bilbiothecaris en senior-assistent voor het [[Library of Congress]].
Gill werd later professor [[zoölogie]] aan de [[George Washington-universiteit]]. Hij was tevens lid van de [[Megatherium Club]] van het [[Smithsonian Institution]]. Hier werd hij door zijn medeleden vaak bespot vanwege zijn ijdelheid. In 1897 werd Gill president van de [[American Association for the Advancement of Science]] in 1897.
